第十五屆中國（）檢驗醫(yī)學(xué)暨輸血儀器試劑博覽會健侖體驗

如果 培養(yǎng)液與細胞的容積比例大于:，生長物質(zhì)彌 散到細胞外的比例將 是細胞內(nèi)達到低于zui小限度的濃度，此時細胞 不能再增殖，培養(yǎng) 液中的pH變堿，抑制細胞生長，細胞變圓不能貼 壁，甚至引起細 胞死亡等。如果接種密度太高，每個細胞周圍培養(yǎng) 液的容積降 至.mm以下，由于彌散率的降低，細胞內(nèi)生物質(zhì) 的濃度增加，容 易使排除物或其他代謝產(chǎn)物達到飽和，能量來源也 很快耗盡，因 此也會妨礙細胞的增殖。如何選擇適宜的接種濃度要根據(jù)細胞的代 謝和生長繁殖速度以及工 作的需要而確定。一般來說，細胞代謝 旺盛、生長速度快的細胞如 腫瘤細胞，接種濃度宜偏低；而正常 組織細胞生長較慢，代謝不夠 旺盛，接種濃度可偏高；如果是為 了保種或不需急用，接種濃度可 偏低些；有時為了便于觀察細胞 形態(tài)結(jié)構(gòu)，接種濃度也可適當(dāng)降低 。七、容器轉(zhuǎn)動速度和懸浮攪 動速度有時為了研究的需要而將培養(yǎng)細胞在容器中進行旋轉(zhuǎn)或攪動 ，如貼 壁細胞在旋轉(zhuǎn)管中培養(yǎng)，使轉(zhuǎn)鼓的轉(zhuǎn)速提高，細胞增殖率 隨之提高 。
